چکیده

The immunochemical technique, first employed in the study of albumin synthesis in chicken liver slices by Peters and Anfmsen (3), has recently been successfully used with rat liver slices by Marsh and Drabkin (4) as well as by Campbell and Stone (5). The present work extends this technique to the problem of the biosynthesis of plasma lipoprotein by rat liver. The liver has been shown to be the site of plasma cholesterol and phospholipid synthesis (6,7). Miller et al. (8) observed that the plasma proteins (exclusive of gamma globulins), were labeled when radioactive amino acids were perfused through the isolated rat liver. Indirect evidence that the liver is capable of synthesizing plasma lipoproteins has recently been obtained by Radding et al. (9) who observed the incorporation of radioactive amino acids into lipoproteins of serum that had been incubated with rat liver slices. The present experiments demonstrate by the immunochemical method a net synthesis of low density lipoproteins in rat liver slices and in the isolated liver perfused with washed red blood cell suspensions. By this means, also, a direct estimate of the rate of synthesis of the cholesterol moiety of the plasma lipoproteins has been obtained for the first time in a mammalian system in vitro.

Evidence for Histidine Residues on Plasma Membrane Phosphatidate Phosphohydrolase from Rat Liver

Objective(s) Phosphatidate phosphohydrolase (PAP) catalyzes the dephosphorylation of phosphatidic acid to yield Pi and  diacylglycerol. Two different forms of PAP in rat hepatocyte have been reported. PAP1 is located in cytosolic and microsomal fractions and participates in the synthesis of triacylglycerols, phosphatidylcholine, and phosphatidylethanolamine, whereas the other form of phosphati...

Studies on Eel Liver Functions Using Perfused Liver and Primary Cultured Hepatocytes

Perfused eel livers, isolated eel hepatocytes, and cultured eel hepatocytes are used to investigate eel liver functions such as gluconeogenesis, glycogen synthesis, and lipoprotein synthesis and methods for preparation are described. A novel phosphoenolpyruvate (PEP) synthesis pathway from pyruvate in gluconeogenesis in eel liver was elucidated and PEP synthesis pathways in eel, rat, and pigeon...

Anti-hypercholesterolemic Effect of Berbamine Isolated from Rhizoma Coptidis in Hypercholesterolemic Zebrafish Induced by High-Cholesterol Diet

The anti-hypercholesterolemic effect of berbamine (BBM) isolated from Rhizoma Coptidis (RC) was investigated in hypercholesterolemic zebrafish model induced by high-cholesterol (HC) diet. Zebrafish embryo assay revealed no significant difference in morphology and cell death with the treatment of BBM less than 20 μg/mL. In zebrafish larvae, the fluorescently labeled cholesterol in caudal artery ...

Total Inhibition of Hepatic P-Lipoprotein Production in the Rat by Orotic Acid*

Lipoproteins of rat plasma have been characterized by lipid analysis, preparative ultracentrifugation, electrophoresis on paper, and immunoelectrophoresis with specific antisera to Lu-lipoprotein, P-lipoprotein, and whole rat plasma. Similar techniques were used to characterize the lipoproteins released by livers perfused in situ with a suspension of erythrocytes in a medium initially free of r...
